Ectopic Expression of PgF3′5′H in Commercial Gypsophila paniculata Cultivar through Optimized Agrobacterium-Mediated Transformation

Gypsophila paniculata is one of the most popular cut flowers in world whose major cultivars are blooming white. As well known, blue could be generated via overexpression gene encoding flavonoid 3′5′-hydroxylase (F3′5′H) species that naturally lack it. In this study, we established regeneration and a genetic transformation system for commercial cultivar ‘YX4’ G. introduced F3′5′H Platycodon grandiflorus (PgF3′5′H) successfully into using protocol. A total 281 hygromycin (Hyg)-resistant plantlets were obtained, 38 them polymerase chain reaction (PCR) positive, indicating 13.5% efficiency. Shoot apex without meristem was more suitable explant due to its high capacity, supplement thidiazuron (TDZ) provided efficient promotion adventitious bud induction, whereas 6-Benzyladenine (6-BA) 1-naphthaleneacetic acid (NAA) did not affect much. Additionally, combination 1 day (d) pre-culture, 5 d co-culture, 10 min infection, 30 mg·L−1 additional acetosyringone (AS) supplement, Hyg selection formed optimized transformation. This reliable agrobacterium-mediated valuable will contribute only creation improvement cultivars, but also function research genes associated with important ornamental traits.
